Does SQL Server 2005 SP2 Contains SP1 updates too or only the SP2 updates.
same is the query for SQL 2000. Does SQL 2000 SP4 contains SP 1,2,3 & 3a ?

Yes, it should contain the updates and fixes for all the previous updates, although it may drop some support for specific things such as in older SP1's. You may wish to look at the change logs for each service pack to ensure they support what you need, and know what features were dropped or not.

Very often, if a service pack (SP) does not contain older SPs, microsoft tend to require older SPs to be installed before, so if it don't require it, it usually contains all older as well. (with nullw0rms exception that if some functionality was dropped, but that's usually no big deal)
